Target Information
This gene generates several transcript variants differing in their first exons. At least three alternatively spliced variants encoding distinct proteins have been reported, two of which encode isoforms functioning as inhibitors of CDK4 kinase. Another transcript includes an alternate first exon located 20 Kb upstream, encoding a structurally unrelated ARF protein that stabilizes tumor suppressor p53 by interacting with and sequestering MDM2. Despite structural and functional differences, both CDK inhibitor isoforms and ARF share a common role in cell cycle G1 control via CDK4 and p53 regulation. This gene is frequently mutated or deleted in various tumors and is recognized as an important tumor suppressor gene.
