Target Information
CD20 is a non-glycosylated surface phosphoprotein with a molecular weight range of 33–37 kDa depending on phosphorylation. It is expressed on mature and most malignant B cells, as well as on a subpopulation of T lymphocytes and follicular dendritic cells. CD20 expression coincides with surface IgM expression and regulates calcium conductance, cell cycle progression, and B-cell proliferation. It associates with lipid rafts in a trigger-dependent manner and remains on the cell surface after receptor ligation. CD20 is a key target for antibody-mediated B-cell depletion therapies, as it is stably expressed on most B-cell malignancies and is not internalized or shed after antibody binding. Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
