Target Information

Focal Adhesion Kinase (FAK) is a 125 kDa non-receptor protein tyrosine kinase that serves as a substrate for Src and plays a key role in integrin signaling. It is involved in cell spreading, differentiation, migration, apoptosis, and the G1–S phase transition. FAK localizes to focal adhesions, which mediate cell attachment to the extracellular matrix via integrin receptors. Tyrosine phosphorylation of FAK increases upon integrin engagement with fibronectin. In murine NIH3T3 fibroblasts, adhesion to fibronectin promotes association of Grb2 and c-Src with FAK, activating ERK2 MAP kinase. Tyr925 has been identified as a critical phosphorylation site for Grb2 SH2 domain binding. Multiple transcript variants of FAK exist, with three full-length isoforms characterized.
