Target Information
LATS2 is a serine/threonine protein kinase belonging to the LATS tumor suppressor family. It interacts with a negative regulator of p53 and functions in a positive feedback loop with p53 that responds to cytoskeleton damage, leading to centrosome and mitotic apparatus dysfunction.
LATS2 plays an essential role in maintaining mitotic fidelity and genomic integrity. It regulates organ size and tumor suppression by restricting proliferation and promoting apoptosis.
The pathway involves a kinase cascade where STK3/MST2 and STK4/MST1, with SAV1, phosphorylate and activate LATS1/2, which then phosphorylate and inactivate YAP1 and WWTR1/TAZ. Phosphorylation of YAP1 by LATS2 inhibits its nuclear translocation, thereby regulating genes involved in cell proliferation, death, and migration.
LATS2 negatively regulates the G1/S transition by down-regulating cyclin E/CDK2 kinase activity.
