MAP4K4 (HGK)는 serine/threonine kinase로서 스트레스 활성화 MAPK 신호 전달 경로, 환경 스트레스 및 TNF-alpha와 같은 cytokine 반응에 중요한 역할을 합니다. JUN N-terminal pathway 상류에서 작용하며, adapter protein Nck의 SH3 도메인과 상호작용하는 것으로 알려져 있습니다. CNH 조절 도메인을 통해 SPG3A N-말단 영역에 결합합니다. 모든 조직에서 발현되며, isoform 5는 뇌에서 풍부하고 isoform 4는 간, 골격근 및 태반에서 주로 발현됩니다.
