Target Information
MERTK (cMER)는 티로신 키나아제 원암유전자로, 망막 색소상피(RPE) 식세포 작용 경로에 관여하며 인간의 망막 질환과 관련이 있습니다. 세포 생존, 이동, 분화, 그리고 세포사멸세포의 식세포작용(efferocytosis) 등 다양한 생리적 과정을 조절합니다. 리간드 결합 시 MERTK의 세포내 도메인에서 자가인산화가 유도되어 하위 신호전달 단백질의 도킹 부위를 제공합니다. 활성화 후 GRB2 또는 PLCG2와 상호작용하며 MAPK1, MAPK2, FAK/PTK2 또는 RAC1의 인산화를 유도합니다. 또한 대식세포의 세포사멸세포 제거, 혈소판 응집, 세포골격 재조직화 등에 관여합니다. RPE에서는 광수용체 외절편의 식세포작용을 조절하며, STAT1을 활성화하여 SOCS1 및 SOCS3의 발현을 유도함으로써 TLR 매개 선천면역반응을 억제하는 역할을 합니다.
