Target Information

Leptin is a 167 amino acid long protein hormone with important effects in regulating body weight, metabolism, and reproductive function. It has a molecular mass of approximately 16 kDa and is encoded by the obese (ob) gene. Leptin is mainly expressed by adipocytes, reflecting its role in sensing total body fat mass. Smaller amounts are secreted by stomach epithelial cells and the placenta.

Leptin receptors are highly expressed in hypothalamic regions regulating body weight, and also found in T lymphocytes and vascular endothelial cells. Leptin influences feeding behavior, hunger, body temperature, and energy expenditure via hypothalamic pathways. It also contributes to hematopoiesis, angiogenesis, wound healing, inflammation, immune responses, pubertal development, and fetal growth. Studies have linked leptin to obesity, anorexia nervosa, insulin resistance, and hypertension. Leptin exhibits thermogenic actions and regulates fatty acid oxidation enzymes. Defects in leptin production can cause severe hereditary obesity in rodents and humans.
