Product Specific Information

Description:
The eBio124-1D1 monoclonal antibody reacts with human CD7 (gp40, Leu9), a 40 kDa receptor belonging to the immunoglobulin gene superfamily. The N-terminal region (aa1–107) is homologous to Ig kappa light chains, while the C-terminal extracellular domain is proline-rich and forms a stalk structure.
CD7 is expressed on most immature and mature T lymphocytes, T cell leukemias, NK cells, a subset of normal B cells, and malignant B cells. Cross-linking of CD7 modulates T and NK cell activity, influencing calcium flux, adhesion molecule expression, cytokine secretion, and proliferation. CD7 associates with phosphoinositol 3′-kinase, and ligation induces D-3 phosphoinositide production and tyrosine phosphorylation.

A subpopulation of human CD34(+) CD38(-) cord blood cells expressing high levels of CD7 has lymphopoietic potential, generating B cells, NK cells, and dendritic cells but not myeloid or erythroid lineages. The CD7(+) phenotype distinguishes primitive lymphoid progenitors from pluripotent stem cells.
CD7 may also cooperate with CD28 in Treg function, as double-deficient mice show reduced Treg numbers and suppressive activity.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells.
Use at 5 µL (0.25 µg) per test (defined as the amount required to stain a 100 µL cell sample).
Typical cell number: 10⁵–10⁸ cells/test.
Excitation: 488 nm
Emission: 520 nm
Laser: Blue Laser
Filtration: 0.2 µm post-manufacturing filtered

Target Information

CD7 (gp40, Leu9) is a 40 kDa member of the immunoglobulin gene superfamily. Its N-terminal domain (aa1–107) is homologous to Ig kappa light chains, while the C-terminal region is proline-rich. CD7 is found on most immature and mature T lymphocytes, T cell leukemias, NK cells, a subset of normal B cells, and malignant B cells.
Cross-linking of surface CD7 modulates T and NK cell activity, enhancing calcium flux, adhesion molecule expression, cytokine secretion, and proliferation. CD7 directly associates with phosphoinositol 3′-kinase, and ligation induces D-3 phosphoinositide production and tyrosine phosphorylation.
CD7 expression is an important marker in leukemia diagnostics.
