Description:
The eBio341 monoclonal antibody reacts with rat CD8β, a member of the immunoglobulin superfamily that heterodimerizes with CD8α to form the CD8 antigen. This antigen is expressed on CD4⁺CD8⁺ double-positive thymocytes, most MHC class I-restricted peripheral T cells, and some macrophages. CD8β expression is absent in athymic mice, indicating dependence on thymic T-cell development. CD8 binds to MHC class I molecules on antigen-presenting cells, aiding T-cell receptor signaling via association with kinase Lck.

Applications Reported: Flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of rat splenocytes and thymocytes. Use ≤0.5 µg per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize antibody titration for best assay performance.

Target Information

The CD8B antigen is a cell surface glycoprotein found on most cytotoxic T lymphocytes. It mediates cell-cell interactions and functions as a coreceptor with the T-cell receptor to recognize antigens presented by MHC class I molecules. The CD8 complex may form as a homodimer (αα) or heterodimer (αβ), both homologous to immunoglobulin variable light chains. The CD8B gene encodes the β chain isoforms, with multiple alternatively spliced variants and a pseudogene located on chromosome 2.
