Target Information

Inositol 1,4,5-trisphosphate (IP3) acts as a second messenger for many growth factors, hormones, and neurotransmitters. Upon binding to IP3 receptor (IP3R), IP3 triggers the release of intracellular calcium into the cytosol. Functional IP3R forms homo- or heterotetramers of ~240 kDa glycoprotein subunits. IP3R is structurally and functionally related to the ryanodine receptor, another intracellular calcium-release channel.

The similarity between IP3R and ryanodine receptor extends to their association with the immunophilin protein FKBP12. Phosphorylation of IP3R by PKC increases IP3-mediated calcium release, while dephosphorylation by calcineurin reduces calcium release, resulting in calcium oscillations. Mammalian IP3R subunits are encoded by three distinct genes with differential expression patterns:

  • IP3R-I: heart, liver, kidney, ovary, and Purkinje neurons
  • IP3R-II: predominantly in brain
  • IP3R-III: pancreatic islets, kidney, and gastrointestinal tract
