Product Specific Information
This antibody detects endogenous levels of IKK alpha/beta only when phosphorylated at Serine 180/181.
Target Information
IKK alpha/beta is encoded by CHUK (conserved helix-loop-helix ubiquitous kinase) and IKKB (inhibitor of kappa light polypeptide gene enhancer in B-cells) located on chromosomes 10 and 8 in humans.
Upon activation of the NF-κB pathway (e.g., by TNF alpha), IKK alpha and IKK beta, together with the regulator NEMO (IKK gamma), phosphorylate IκB proteins. This leads to their ubiquitin-mediated degradation and the subsequent activation and nuclear translocation of NF-κB.
Phosphorylation at serine 176 and 180 in the activation loop induces conformational changes required for kinase activation.
