Target Information
Protein kinases are enzymes that transfer a phosphate group from a phosphate donor, generally the γ phosphate of ATP, onto an acceptor amino acid in a substrate protein. Through this mechanism, protein kinases mediate most signal transduction in eukaryotic cells, regulating cellular metabolism, transcription, cell cycle progression, cytoskeletal rearrangement, cell movement, apoptosis, and differentiation.
With more than 500 gene products, the protein kinase family is one of the largest in eukaryotes. It is classified into eight major groups based on catalytic domain sequence comparison (tyrosine or serine/threonine kinases). The AGC kinase group includes 63 kinases, such as:
- Cyclic nucleotide-regulated protein kinases (PKA & PKG)
- Diacylglycerol-activated/phospholipid-dependent protein kinase C (PKC)
- RAC/Akt protein kinase family
- G protein-coupled receptor kinases (ARK)
- Ribosomal protein S6 kinases (RSK)
