Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation. p53 up-regulates transcription of genes such as p21waf1, leading to G1 or G2/M arrest. Structurally, p53 includes an N-terminal transactivation domain, a central DNA-binding domain, an oligomerisation domain, and a C-terminal regulatory domain.

Phosphorylation at Ser15 is critical for activation and stabilization, while phosphorylation at Ser392 is associated with human tumor formation. p53 plays a role in preventing proliferation of damaged cells and functions as an anticancer factor. Its expression increases under stress and aging conditions, and elevated levels are linked to deficits in long-term potentiation (LTP) related to learning and memory.

Mutations in the TP53 gene are common in various human cancers and lead to loss of tumor suppressor activity. Germline mutations can also occur in cancer-prone families, such as those with Li-Fraumeni syndrome.
