Target Information
Thymidylate Synthase (TS) catalyzes the conversion of deoxyuridine monophosphate (dUMP) to deoxythymidine monophosphate (dTMP), a key step in DNA biosynthesis. TS is a critical target of fluoropyrimidines, an important class of antineoplastic drugs used to treat various solid tumors. Both 5-fluorouracil (5-FU) and fluorodeoxyuridine are converted in tumor cells to FdUMP, which inactivates TS through formation of a ternary complex with the folate cofactor 5,10-methylenetetrahydrofolate. Expression of TS protein correlates with response to 5-FU in human colorectal, gastric, head and neck, and breast carcinomas.
