Target Information
IRS-1, a major substrate of the insulin receptor, is phosphorylated in response to stimulation by insulin, insulin-like growth factor 1 (IGF-1), and interleukin 4 (IL-4).
IRS-1 is phosphorylated on serine, threonine, and tyrosine residues in various tissues. Casein kinase II mediates part of the insulin-stimulated serine/threonine phosphorylation of IRS-1 in vivo.
Thr502 and Ser99 are identified phosphorylation sites catalyzed by casein kinase II. This phosphorylation may be a component of the intracellular insulin signaling cascade.
IRS-1 also contains three putative 14-3-3 binding sites (Ser270, Ser374, Ser641), with the motif around Ser270 located in the phosphotyrosine-binding domain responsible for interaction with the insulin receptor.
