This product is diluted and ready-to-use.
Recommended positive control tissue: cerebellum (not limited to this tissue type).
Intended for laboratory professional use in detecting the corresponding protein in formalin-fixed, paraffin-embedded tissue stained by manual qualitative IHC testing.
Recognizes a protein of approximately 50 kDa identified as gamma-enolase.
Three enolase isoenzymes (α, β, γ) exist:
- α-isoform: expressed in most tissues
- β-isoform: predominantly in muscle
- γ-enolase: found in nervous tissue
These isoforms function in glycolysis, converting phosphoglyceric acid to phosphoenolpyruvic acid.
NSE-gamma serves as a marker for peripheral nerves and neuroendocrine tumors such as pheochromocytomas, often used with synaptophysin, Chromogranin A, and Neurofilament.
Pretreatment of deparaffinized tissue with heat-induced or enzymatic epitope retrieval is recommended.
IHC staining visualizes antigens via sequential antibody application and chromogenic substrate reaction.
Results are interpreted under a light microscope for differential diagnosis.
A positive tissue control must accompany each staining procedure.
External positive controls should be freshly prepared and processed similarly to patient samples.
Weakly positive tissues are preferred for quality control to detect minor reagent degradation.
Negative control tissues should demonstrate absence of specific staining.
Target Information
Neuron Specific Enolase (NSE, ENO1, ENO2, ENO3) catalyzes the conversion of 2-phosphoglycerate to phosphoenolpyruvate in glycolysis and the reverse in gluconeogenesis.
NSE is stable in biological fluids and diffuses to CSF upon neuronal injury.
- α-subunit: expressed in most tissues
- β-subunit: muscle-specific
- γ-subunit: neuron-specific
NSE co-expresses with Chromogranin A in neuroendocrine neoplasms.
Antibodies to NSE are useful for identifying neuronal cell bodies, neuroendocrine cells, and neuronal lineage.
NSE release into CSF and blood serves as a biomarker for neuronal injury and is clinically used in conditions such as stroke, hypoxic brain damage, status epilepticus, Creutzfeldt-Jakob disease, and herpetic encephalitis.
Elevated NSE levels are found in neuroblastoma and small cell lung cancer.
