For reconstitution, add 100 µL distilled water to achieve a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments, perform an additional desalting step using Zeba Spin Desalting Columns (7K MWCO, 0.5 mL, Product #89882).

Target Information

Modification of target proteins by ubiquitin participates in a wide array of biological functions. Proteins destined for degradation or processing via the 26S proteasome are coupled to multiple copies of ubiquitin. Attachment of ubiquitin or ubiquitin-related molecules may also alter subcellular distribution or protein activity.

Deubiquitination, catalyzed by deubiquitinating enzymes, provides another level of ubiquitin regulation. These enzymes fall into four families:

1. Ubiquitin C-terminal hydrolases
2. Ubiquitin-specific processing proteases (USPs)
3. OTU-domain ubiquitin-aldehyde-binding proteins
4. Jab1/Pad1/MPN-domain-containing metallo-enzymes

Among these, USPs are the most widespread and evolutionarily conserved, releasing ubiquitin from conjugated proteins. They possess conserved catalytic domains with Cys and His boxes, and variable N- or C-terminal extensions that mediate substrate recognition, localization, and protein interactions.
