Target Information
RIPK1은 세포사멸 및 염증 신호 전달에 관여하는 세린/트레오닌 키나아제로, death receptor 결합, 병원체 인식 수용체(PRRs) 활성화, DNA 손상 등에 의해 활성화됩니다. TNF-α에 의해 TNFR1이 활성화되면 TRADD와 TRAF2가 수용체에 결합하고, RIPK1은 DAB2IP를 Ser-728에서 인산화하여 MAP3K5-JNK 경로를 활성화합니다. TRAF2에 의한 Lys-63 연쇄 유비퀴틴화는 NF-κB 및 MAPK 경로의 신호 전달을 강화합니다. 또한 RIPK1과 RIPK3의 상호작용은 괴사(necrosis)를 유도하는 복합체를 형성합니다.
