Target Information
RIPK1은 세포사멸 및 염증 신호를 전달하는 세린-트레오닌 키나아제로, 사멸 수용체 결합, 병원체 인식 수용체(PRRs) 활성화, DNA 손상에 의해 활성화된다. TNFR1이 TNF-α에 의해 활성화되면 TRADD 및 TRAF2가 수용체에 모집된다. RIPK1은 DAB2IP를 Ser-728에서 인산화하여 MAP3K5-JNK 세포사멸 경로를 활성화한다. TRAF2에 의한 Lys-63 결합형 폴리유비퀴틴화는 MAPK 및 NF-κB 경로에서 하위 신호전달을 강화한다. 폴리유비퀴틴화된 RIPK1은 IKBKG/NEMO와 결합하여 NF-κB 활성화를 유도하며, RIPK3와 결합 시 괴사 유도 복합체를 형성한다.
