MHC class II molecules are heterodimers composed of non-covalently associated alpha (31–34 kDa) and beta (26–29 kDa) chains.
These molecules present exogenously derived antigens to CD4⁺ T lymphocytes (T helper cells).
Antigen presentation involves lysosomal proteases that degrade both endocytosed antigens and the invariant chain (Ii), enabling peptide binding.
CD4 interacts with non-polymorphic residues of MHC class II.
HLA and MHC antibodies are essential tools in immunopeptidomics, aiding in the identification and characterization of neoantigens via high-performance liquid chromatography and tandem mass spectrometry.
