Background
Mammalian mitochondrial ribosomal proteins are encoded by nuclear genes and are essential for protein synthesis within the mitochondrion. Mitochondrial ribosomes (mitoribosomes) consist of a small 28S subunit and a large 39S subunit, with approximately 75% protein to rRNA composition—opposite to prokaryotic ribosomes. Unlike prokaryotic ribosomes, mammalian mitoribosomes lack 5S rRNA. The proteins composing the mitoribosome vary significantly among species, making sequence homology recognition difficult.
The MRPS22 gene encodes a 28S subunit protein unique to mammalian mitochondria, with no counterpart in prokaryotic or fungal mitochondrial ribosomes. This gene is located telomeric to and transcribed in the opposite direction from the forkhead box L2 gene. A pseudogene also exists.
