Background
Glutamate metabotropic receptor 4 (GRM4) is a G protein-coupled receptor expressed in the central nervous system. L-glutamate, the major excitatory neurotransmitter, activates both ionotropic and metabotropic glutamate receptors. Glutamatergic neurotransmission is essential for normal brain function and is implicated in various neuropathologic conditions.
Metabotropic glutamate receptors are classified into three groups based on sequence homology, signaling mechanisms, and pharmacologic properties:
- Group I: GRM1, GRM5 (activate phospholipase C)
- Group II: GRM2, GRM3
- Group III: GRM4, GRM6, GRM7, GRM8 (linked to inhibition of the cyclic AMP cascade, differing in agonist selectivity)
Several transcript variants encoding different isoforms have been identified for this gene.
