Background

  • Catalytic activity: S-adenosyl-L-methionine + histone L-lysine → S-adenosyl-L-homocysteine + histone N(6)-methyl-L-lysine
  • Domain: SET domain contains the active site; pre-SET and post-SET domains are required for methyltransferase activity and stable heterochromatin binding.
  • Function: Histone methyltransferase that specifically trimethylates Lys-9 of histone H3, marking epigenetic transcriptional repression by recruiting HP1 proteins (CBX1, CBX3, CBX5). Functions in heterochromatin regions, crucial for constitutive heterochromatin formation at pericentric and telomeric regions. Also directs DNA methylation at pericentric repeats.
  • Similarity: Belongs to the histone-lysine methyltransferase family, Suvar3-9 subfamily; contains chromo, pre-SET, post-SET, and SET domains.
