Product Specific Information

Description:
The DX5 monoclonal antibody reacts with CD49b, an antigen expressed on a majority of mouse natural killer (NK) cells and a subset of T cells. DX5 reacts with all tested mouse strains including BALB/c, C57BL/6, C3H, CBA, DBA, AKR, SJL, and 129. Simultaneous staining with anti-NK1.1 mAb (PK136) shows coexpression of both markers by most NK cells, with minor DX5⁺PK136⁻ and DX5⁻PK136⁺ populations.

Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.5 µg/test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test; titration recommended for optimal performance.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation; protect vial and samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-822-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be empirically verified.

Excitation / Emission / Laser:
488–561 nm excitation; 775 nm emission; compatible with Blue, Green, Yellow-Green lasers.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

CD49b (Integrin alpha 2) is a member of the integrin family. It forms a heterodimer with CD29 (Integrin beta 1) to create the VLA-2 (Integrin α2β1) complex, functioning as a receptor for laminin, collagen, and fibronectin. It plays roles in VEGF-induced angiogenesis, adhesion, and lymphocyte activation.
CD49b is expressed by NK cells, NK-T cells, monocytes, platelets, epithelial cells, and subsets of T and B cells, including CD4⁺ Tr1 regulatory cells co-expressing LAG-3.
