Guanylate kinase (GK) catalyzes the ATP-dependent phosphorylation of GMP into GDP and is essential for recycling GMP and indirectly cGMP.
GK is a highly conserved monomeric protein (~200 amino acids) found in prokaryotes, lower eukaryotes, and vertebrates.
It shares structural similarity with Vaccinia virus protein A57R (SalG2R).
Proteins with DHR, SH3, and GK-like domains are termed MAGUKs (membrane-associated guanylate kinase homologs), including:
- Drosophila lethal(1)discs large-1 tumor suppressor protein (dlg1)
- Mammalian tight junction protein Zo-1
- Synaptic proteins interacting with NMDA receptor subunits (SAP90/PSD-95, CHAPSYN-110/PSD-93, SAP97/DLG1, SAP102)
- Vertebrate erythrocyte membrane protein p55
- C. elegans lin-2
- Rat protein CASK
- Human DLG2, DLG3
The ATP-binding site (P-loop) in GK’s N-terminal region is not conserved in GK-like domains of MAGUKs, but these proteins retain residues involved in MP binding.
