Target Information
Huntingtin은 헌팅턴병(Huntington’s disease, HD)과 관련된 유전자 산물로, 신경퇴행성 질환인 헌팅턴병은 선조체 신경세포의 손실로 특징지어집니다. 이 질환은 헌팅틴 유전자 내 불안정한 삼염기 반복 확장에 의해 발생하며, 단백질 내 폴리글루타민 반복을 유발합니다.
HD는 중년기에 발병하는 상염색체 우성 신경퇴행성 질환으로, 정신질환, 치매, 불수의 운동(무도증)을 동반하며 10–20년 내 사망에 이릅니다. 헌팅틴 유전자는 약 180 kb 크기로 67개의 엑손을 포함하고 있으며, 정상 발달에 필수적입니다.
두 가지 형태의 전사체가 존재하며, 각각 13.7 kb 및 10.3 kb 크기로 다양한 태아 및 성체 조직에서 발현됩니다. 유전적 결함은 전사를 완전히 억제하지는 않지만 mRNA의 새로운 특성이나 단백질 기능 변화를 유발할 수 있습니다.
