Product Specific Information

The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein. CD40 is a TNFR family member expressed on B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells. It regulates B cell development, maturation, and isotype switching, and promotes proliferation and survival when co-stimulated (e.g., with IL-4). The CD40–CD154 interaction is critical for T–B cell communication and immune regulation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated for flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Light sensitivity: Protect from light due to photo-induced oxidation.
Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54).
Excitation/Emission: 488–561 nm / 775 nm; compatible with Blue, Green, and Yellow-Green lasers.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

The CD40 antigen is a single-chain glycoprotein, part of the TNF/NGF receptor superfamily, and homologous to CD30. It is expressed on all B cells (except plasma cells), as well as some epithelial cells, carcinomas, and lymphoid dendritic cells. CD40 plays a pivotal role in immune responses, including immunoglobulin class switching, memory B cell development, and germinal center formation. It is regulated by AT-hook transcription factor AKNA and interacts with adaptor protein TNFR2 for signal transduction. CD40–CD154 interaction is also implicated in amyloid-beta-induced microglial activation, an early event in Alzheimer’s disease. Mutations in CD40 are associated with Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
