Target Information

TANK was initially identified as a novel TRAF-interacting protein that regulated TRAF-mediated signal transduction. Ligand binding by surface receptors in the tumor necrosis factor (TNF) receptor and Toll/interleukin-1 (IL-1) receptor families leads to the formation of a TRAF/TANK complex that mediates activation of NF-kappa-B through association with kinases IKK-iota and TBK1. These proteins can form a complex with NEMO, regulating Ikappa-B activity. This suggests that TBK1 and IKK-iota may help modulate NF-kappa-B transactivation functions. At least two isoforms of TANK are known.
