Target Information
DR3/TNFRSF25 is a member of the TNF-receptor superfamily, expressed mainly in lymphocyte-rich tissues. It plays a role in lymphocyte homeostasis, NF-kappa B activation, and apoptosis regulation.
Signal transduction involves adaptor proteins containing death domains. Mouse knockout studies suggest its role in removing self-reactive T cells in the thymus.
Multiple alternatively spliced transcript variants exist, encoding distinct isoforms, many of which are secreted. In activated B and T cells, splicing shifts toward producing full-length, membrane-bound isoforms, implicated in lymphocyte proliferation control.
