Product Specific Information

The 2C9.G3 (HMb3-1) monoclonal antibody reacts with mouse and rat CD61 (integrin beta3), expressed by activated T cells, granulocytes, and platelets. CD61 associates non-covalently with alphaV (CD51) to form Vitronectin Receptor and with alphaIIb (CD41) to form gpIIb/IIIa, mediating adhesion to extracellular matrix components such as fibrinogen, fibronectin, vitronectin, thrombospondin, and von Willebrand factor.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes and bone marrow cells at ≤ 0.05 µg/test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Titrate for optimal performance.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.
  • Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample or per 100 µL of cell sample for single-color controls.
  • NovaFluor conjugates utilize Phiton™ technology, providing engineered fluorescent signatures optimized for minimal spillover and spread.

Target Information

CD61 (GPIIIa, ITGB3) is a 105 kDa glycoprotein found on megakaryocytes, platelets, and their precursors. It functions as a receptor for fibrinogen, fibronectin, von Willebrand factor, and vitronectin, forming complexes with CD41 or CD51. CD61 is involved in platelet aggregation, cell adhesion, and signaling. Dysfunctions are associated with Glanzmann Thrombasthenia and Platelet type-16 Bleeding Disorder.
