Product Specific Information

The 2C9.G3 (HMb3-1) monoclonal antibody reacts with mouse and rat CD61 (integrin beta3). CD61 is expressed by activated T cells, granulocytes, and platelets. It forms heterodimeric complexes with integrin alphaV (CD51) and alphaIIb (CD41), responsible for adhesion to extracellular matrix components such as fibrinogen, fibronectin, vitronectin, thrombospondin, and von Willebrand factor.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Mouse splenocytes and bone marrow cells (≤ 0.1 µg/test). Recommended careful titration for optimal assay performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures considering spillover and spread impacts.
Excitation: 552 nm; Emission: 718 nm; Laser: 561 nm (Yellow).

Internal testing shows NovaFluor Yellow 730 may non-specifically stain B cells in SJL mice, but not in BALB/c or C57BL/6 strains.

Target Information

CD61 (GPIIIa, ITGB3) is a 105 kDa glycoprotein found on megakaryocytes, platelets, and their precursors. It acts as a receptor for fibrinogen, fibronectin, von Willebrand factor, and vitronectin. CD61 associates with alphaIIb (CD41) or alphaV (CD51) integrins and participates in cell adhesion and cell-surface mediated signaling.
Diseases associated with CD61 dysfunction include Glanzmann Thrombasthenia and Platelet type-16 Bleeding Disorder.
