Target Information
Juvenile nephronophthisis type 1 is caused by mutations of NPHP1, the gene encoding for nephrocystin.
Nephrocystin interacts with p130Cas (BCAR1), PTK2B (Pyk2), and tensin in embryonic kidney and testis, suggesting a shared signaling pathway.
In mammalian cells, nephrocystin and p130Cas localize at cell-cell contact sites in polarized Madin-Darby canine kidney epithelial cells.
Expression of nephrocystin induces phosphorylation of Pyk2 on Tyrosine 402 and activates downstream MAP kinases (ERK1, ERK2).
Nephrocystin contains a conserved src-homology 3 (SH3) domain.
