Target Information

Huntington’s disease (HD) is a neurodegenerative disorder characterized by loss of striatal neurons, caused by an expansion of a polyglutamine tract in the huntingtin protein. HAP1 (Huntingtin-associated protein 1) was identified through two-hybrid library screening; its binding to huntingtin correlates with polyglutamine tract expansion.
HAP1 interacts with cytoskeletal proteins such as dynactin and pericentriolar autoantigen protein 1, indicating a role in vesicular trafficking or organelle transport. It is also involved in huntingtin-enhanced BDNF transport along microtubules. Impaired function of this process leads to reduced neurotrophic support and neuronal toxicity, potentially contributing to disease pathogenesis.
Several alternatively spliced isoforms of HAP1 have been described.
