Target Information
Ago proteins are broadly expressed in somatic cells and associate with miRNAs, acting as key mediators in RNA silencing pathways. Only the Ago2 protein exhibits endonucleolytic “Slicer” activity, enabling miRNA-directed cleavage of target mRNA when base-pairing is perfect. With partial complementarity, Ago2 represses translation instead of cleavage. It plays essential roles in RNA interference, translation repression, and heterochromatin formation as a core component of the RISC complex. Ago2 is also involved in siRNA/miRNA-guided gene silencing, RNA slicing independent of Dicer, and regulation of miRNA maturation. Overexpression of Ago2 has been linked to tumor growth and survival, and gene disruption studies in mice show its necessity for embryonic development and B-lymphoid and erythroid regulation.
