The Human CoV-NL63 Ig Total Simplex ProcartaPlex Kit measures anti-CoV-NL63 antibodies in serum and plasma. The detector antibody is an Immunoglobulin (Ig) Total antibody directly conjugated to phycoerythrin (PE) that cross-reacts with all Ig (IgG, IgM, and IgA) in serum and plasma. IgG is the most abundant of the immunoglobulins.

This kit is designed to be combinable with other Simplex kits from the Coronavirus Panel so that you can create your own multiplex panel utilizing Luminex xMAP technology for protein detection and quantitation. When combining multiple Simplex kits (i.e., when not using a pre-configured Multiplex Panel), only one Viral Reagent Kit (sold separately) is required per assay plate regardless of plex size.

About ProcartaPlex Assays for the Luminex Platform

ProcartaPlex immunoassays are based on the sandwich ELISA principle, using two highly specific antibodies binding to different epitopes of one protein to quantitate up to:

• 80 protein targets with FLEXMAP 3D or Luminex 200
• 50 protein targets with MAGPIX

These assays require as little as 25 μL of plasma or serum, or 50 μL of cell culture supernatant, and yield analyzed results in approximately four hours.

Technology Overview

Luminex MagPlex superparamagnetic microsphere beads are internally dyed with precise ratios of red and infrared fluorophores to create 100 spectrally unique bead signatures identifiable by Luminex detection systems (Luminex 200, FLEXMAP 3D, MAGPIX).
Similar to a sandwich ELISA, matched antibody pairs identify the target protein. Each unique bead is labeled with a specific antibody, and bound proteins are detected with biotinylated antibodies and streptavidin–R-phycoerythrin (RPE).

Unlike ELISA, the capture antibody in ProcartaPlex assays is conjugated to a magnetic bead and free-floating in solution. Detection is achieved via dual-laser systems that distinguish bead signatures and quantify RPE fluorescence proportional to protein concentration.

ProcartaPlex multiplex assays can profile up to 80× more targets using less sample and time compared to traditional ELISA.
