The Human SARS S1 Ig Total Simplex ProcartaPlex™ Kit measures anti-SARS S1 antibodies in serum and plasma. The detector antibody is an Immunoglobulin (Ig) Total antibody directly conjugated to phycoerythrin (PE) that cross-reacts with all Ig types (IgG, IgM, and IgA) in serum and plasma. IgG is the most abundant of the immunoglobulins.

This kit is designed to be combinable with other Simplex kits from the Coronavirus Panel, allowing you to create your own multiplex panel utilizing Luminex xMAP technology for protein detection and quantitation. When combining multiple Simplex kits (i.e., when not using a pre-configured Multiplex Panel), only one Viral Reagent kit (sold separately) is required per assay plate regardless of plex size.

About ProcartaPlex Assays for the Luminex Platform

ProcartaPlex immunoassays are based on the sandwich ELISA principle, using two highly specific antibodies binding to different epitopes of one protein to quantitate up to:

• 80 protein targets simultaneously with FLEXMAP 3D or Luminex 200 instruments
• 50 protein targets with the MAGPIX instrument

Only 25 μL of plasma or serum, or 50 μL of cell culture supernatant is needed, and results can be obtained in approximately 4 hours.

Technology Overview

The Luminex MagPlex superparamagnetic microsphere beads used in the ProcartaPlex assay are internally dyed with precise proportions of red and infrared fluorophores to create 100 spectrally unique bead signatures identifiable by Luminex xMAP detection systems (Luminex 200, FLEXMAP 3D, and MAGPIX).

Like a sandwich ELISA, the ProcartaPlex assay uses matched antibody pairs to identify the protein of interest. Each spectrally unique bead is labeled with antibodies specific for a single target protein, and bound proteins are detected with biotinylated antibodies and streptavidin–R-phycoerythrin (RPE). The conjugation of antibodies to distinct beads enables simultaneous analysis of multiple targets in a single well.

Unlike traditional ELISA, the capture antibody in ProcartaPlex assays is conjugated to a magnetic bead rather than adsorbed to a microplate well, allowing reagents to remain free-floating in solution. The Luminex 200 instrument uses two lasers—one to distinguish bead signatures and another to quantify RPE fluorescence, which is proportional to the protein concentration. ProcartaPlex multiplex assays can profile up to 80 targets using significantly less sample and time compared to a standard ELISA.

ProcartaPlex Simplex kits offer the flexibility to create unique panels, with over 90% of targets combinable. Kits are available for six species: human, mouse, rat, nonhuman primate, porcine, and canine.
