Product Specific Information

Mouse and rat FAK-1 proteins share 93% homology with human. On western blots, this antibody identifies the target band at approximately 110 kDa. Reactivity has been confirmed with HeLa cell lysates and mouse and rat brain homogenates by western blotting.


Target Information

Focal Adhesion Kinase (FAK) is a 125 kDa non-receptor protein tyrosine kinase acting as a substrate for Src and a key element of integrin signaling. It plays an essential role in cell spreading, differentiation, migration, cell death, and the G1–S phase transition of the cell cycle. FAK localizes to focal adhesions, sites where cells attach to the extracellular matrix via integrin receptors. Increased FAK tyrosine phosphorylation occurs upon integrin engagement with fibronectin. Adhesion of murine NIH3T3 fibroblasts to fibronectin promotes association of Grb2 and c-Src with FAK, resulting in activation of ERK2 MAP kinase. In v-Src-transformed NIH3T3 cells, the association of v-Src, Grb2, and Sos with FAK is independent of adhesion. The Grb2 SH2 domain binds directly to tyrosine-phosphorylated FAK at Tyr925. Several transcript variants encoding different isoforms have been identified for the FAK gene.
