Target Information

LSD1 is a histone demethylase that specifically demethylates Lys-4 of histone H3, a tag for epigenetic transcriptional activation, acting as a corepressor. It contains a SWIRM domain, a FAD-binding motif, and an amine oxidase domain. LSD1 is part of several histone deacetylase complexes and silences genes via demethylation. It oxidizes substrates through FAD, generating imine intermediates that are hydrolyzed. LSD1 demethylates mono- and tri-methylated Lys-4 of histone H3 and may repress neuronal genes. It requires RCOR1/CoREST to demethylate H3 Lys-4 on nucleosomes. LSD1 can also demethylate Lys-9 of histone H3, leading to derepression of androgen receptor target genes. Mutations in this gene can result in cleft palate, psychomotor retardation, and distinctive facial features.
