This antibody detects endogenous levels of total TINF2.
Target Information
TRF1 is a negative regulator of telomere length maintenance. Two proteins, tankyrase and Tin2, which bind to mammalian TRF1, have been identified.
Tankyrase, a protein with homology to ankyrins and the catalytic domain of poly(ADP-ribose) polymerase (PARP), localizes to telomeres by binding to TRF1 through its ankyrin repeats.
Tankyrase exhibits PARP activity and participates in ADP-ribosylation of TRF1, reducing its ability to bind telomeric DNA. This suggests telomere function in human cells is regulated by poly(ADP)-ribosylation.
TIN2, identified as a TRF1-interacting partner, localizes to telomeres and is essential for proper regulation of telomere length. Like TRF1, TIN2 is widely and constitutively expressed, acting to counterbalance telomere elongation by telomerase. However, endogenous TIN2 expression is low, making visualization by immunostaining challenging in some systems.
