Target Information
HIF1-alpha (HIF1A) is a subunit of HIF1, a transcription factor found in mammalian cells under hypoxic conditions. HIF-1 consists of alpha and beta subunits, both belonging to the basic-helix-loop-helix PAS family of transcription factors.
HIF1 functions as a transcriptional regulator of adaptive responses to hypoxia, activating over 40 genes including erythropoietin, glucose transporters, glycolytic enzymes, VEGF, and HILPDA. These genes enhance oxygen delivery and metabolic adaptation under low oxygen conditions.
HIF1-alpha also regulates hypoxia-mediated apoptosis, cell proliferation, and tumor angiogenesis. Under hypoxia, p53 protein accumulation interacts with HIF1-alpha, promoting MDM2-mediated ubiquitination and degradation of HIF1-alpha. Induction of NOX4 by HIF1-alpha helps maintain ROS levels and supports hypoxia-induced proliferation.
In humans, HIF1A is located on chromosome 14q. The C-terminal binds to p300, forming p300/CBP-HIF complexes that induce hypoxia-responsive genes such as VEGF. Hypoxia plays a critical role in diseases like myocardial and cerebral ischemia, cancer, pulmonary hypertension, congenital heart disease, and COPD.
