Target Information
The structure-specific ERCC1/XPF endonuclease complex is involved in the repair of two distinct types of DNA lesions:
- Nucleotide excision repair (NER) for UV-induced lesions and bulky chemical adducts
- Recombination repair of genotoxic interstrand cross-links
NER involves dual incisions on both sides of the DNA damage catalyzed by two nucleases. In mammalian cells, XPG cleaves the 3′ side of the lesion, while ERCC1/XPF makes the 5′ incision. ERCC1/XPF also likely plays a role in processing partially repaired recombination intermediates.
