Target Information
HtrA1 is a 51 kDa protein belonging to the highly conserved mammalian HtrA serine protease family. It is a human homolog of bacterial HtrA and includes:
- Secretory signal sequence
- IGFBP-binding domain
- Kazal-type serine-protease inhibitor domain (N-terminus)
- Trypsin-like serine/protease domain and PDZ domain (C-terminus)
HtrA1 plays a crucial role in cell growth regulation, inhibits TGF-beta signaling, and is involved in various physiological processes. It is up-regulated in the placenta during pregnancy, contributing to implantation and placental physiology. HtrA1 regulates IGF availability by cleaving IGF-binding proteins, acts as a tumor suppressor, and is down-regulated in ovarian cancers and melanomas. Its expression is implicated in osteoarthritis, suggesting therapeutic potential. Northern blot analysis shows high expression in placenta.
