Target Information
Smad4 단백질은 인간 염색체 18에 위치한 Smad4 유전자로부터 발현되며, Smad family member 4, MAD homolog 4 등으로도 알려져 있음.
Smad4는 N-말단 MH1(MAD homology 1)과 C-말단 MH2(MAD homology 2) 도메인을 포함하며, 각각 DNA 결합 및 단백질 상호작용에 관여함.
TGF-beta 및 BMP 등의 리간드 결합 시 Smad1, 2, 3, 5, 8(R-Smad)이 인산화되어 Smad4(co-Smad)와 복합체를 형성하고 핵으로 이동함.
Smad4는 Cyclin D1의 발현 억제 및 콜라겐 발현 증가를 통해 세포 증식을 억제하는 종양 억제 단백질로 작용함.
탈인산화 과정은 Smad 단백질의 핵 내외 이동을 조절함.
