Target Information
TMP21 is a ubiquitously expressed protein involved in vesicular targeting and protein transport. It is a component of the presenilin complex, modulating the γ-secretase but not the ε-secretase cleavage activity of the amyloid precursor protein.
The presenilin complex includes APH1, nicastrin, PEN2, and presenilin-1. These proteins cleave the amyloid precursor protein at γ- and ε-sites, leading to accumulation of the Abeta cleavage product associated with Alzheimer’s disease.
Co-immunoprecipitation experiments have shown TMP21’s involvement in this complex. siRNA-mediated suppression of TMP21 increases γ-secretase activity and Abeta production, indicating TMP21’s regulatory role in this pathway.
