Target Information
PAI-1 (plasminogen activator inhibitor 1)은 세린 프로테아제 억제제(superfamily of serine protease inhibitors)에 속하며, 조직형 및 유로키나제형 플라스미노겐 활성화제(tPA 및 uPA)의 주요 억제제입니다.
혈소판이 주요 순환 PAI-1의 원천이지만, 섬유아세포, 평활근세포, 내피세포, 간세포 및 염증세포 등 다양한 세포 유형에서도 합성 및 분비됩니다.
PAI-1의 발현은 성장인자, 사이토카인, 호르몬, 염증 인자, 대사물질, 혈관 조절 인자 및 환경적 요인 등에 의해 전사 수준에서 조절됩니다.
PAI-1의 과발현은 비만, 혈전성 질환, 대사 증후군 등과 관련된 혈전 발생 증가와 연관이 있으며, 결핍 시 비정상적인 출혈을 유발할 수 있습니다.
또한, PAI-1은 플라스미노겐 활성화제의 활성을 억제하여 섬유소 용해를 억제하고, 신경세포 생존을 촉진할 수 있습니다.
PAI-1 결핍(plasminogen activator inhibitor-1 deficiency)은 다양한 형태의 대체 스플라이싱으로 인해 서로 다른 아이소폼을 생성할 수 있습니다.
