Target Information

Junctional complexes between the plasma membrane (PM) and endoplasmic/sarcoplasmic reticulum (ER/SR) are common in excitable cells and mediate communication between surface and intracellular ion channels.
Junctophilins (JPs) are essential components of these complexes, consisting of a hydrophobic C-terminal segment spanning the ER/SR membrane and a cytoplasmic domain with affinity for the PM.

Four tissue-specific JP subtypes are known:

  • JPH1: Expressed in skeletal muscle
  • JPH2: Present in all muscle cell types
  • JPH3 and JPH4: Predominantly expressed in the brain

In the CNS, JPH3 and JPH4 contribute to subsurface cistern formation in neurons. Mice lacking both subtypes show impaired learning, memory deficits, and abnormal neural functions.
