Recommended positive control tissue: Tonsil (other tissues may also be suitable).
The primary antibody is intended for professional laboratory use in detecting the corresponding protein in formalin-fixed, paraffin-embedded tissues by manual qualitative IHC. It should be applied after tumor diagnosis by conventional histopathology.
Recognizes a 33 kDa glycoprotein, nucleophosmin (NPM), predominantly localized in the nucleus. NPM is involved in ribosomal assembly and rRNA transport, and is highly phosphorylated during mitosis. It shuttles between the nucleus and cytoplasm and participates in ARF/p53 pathway regulation.
Mutations in exon 12 are associated with aberrant cytoplasmic localization and acute myeloid leukemia (AML). This antibody aids in AML classification.
For IHC, use with formalin-fixed, paraffin-embedded sections. Pretreatment with heat-induced or enzymatic antigen retrieval is recommended.
IHC staining allows visualization of antigens through sequential application of primary and secondary antibodies, enzyme complex, and chromogenic substrate. Results are interpreted microscopically for diagnostic support.
A positive tissue control must be run with each staining procedure. Control tissues should include both positive and negative staining elements. External positive controls should be processed identically to patient samples and selected for weak positive expression to detect subtle reagent degradation or method issues.
Negative controls (internal or external) ensure absence of non-specific staining and validate results.
Target Information
NPM1 (Nucleophosmin 1, B23, NO38, Nutramin) is a ubiquitously expressed phosphoprotein involved in:
- Ribosome assembly and transport
- Nucleocytoplasmic trafficking
- Regulation of DNA polymerase α activity
- Centrosome duplication
- Regulation of p53 pathway
NPM1 shuttles between the nucleus, cytoplasm, and nucleolus, chaperoning core histones. It binds nucleic acids, prevents protein aggregation, and assists in protein renaturation.
It participates in tumor-associated fusion proteins such as NPM-ALK, NPM-RAR, and NPM-MLF1, contributing to oncogenic processes.
Dysfunction of NPM1 is linked to acute myeloid leukemia and lymphomatoid papulosis.
