Background

Glutamate decarboxylase 1 (GAD1) is one of several forms of glutamic acid decarboxylase, identified as a major autoantigen in insulin-dependent diabetes. This enzyme catalyzes the conversion of L-glutamic acid to gamma-aminobutyric acid (GABA). It is implicated as an autoantigen and autoreactive T cell target in insulin-dependent diabetes and may also play a role in stiff man syndrome. Deficiency in this enzyme can lead to pyridoxine dependency with seizures. Alternative splicing results in two products: a predominant 67-kDa form and a less-frequent 25-kDa form.
