Cellular Localization Cell membrane; Multi-pass membrane protein; Cell junction; Early endosome; Late endosome; Lysosome. In unstimulated cells, diffuse on plasma membrane; upon agonist stimulation, colocalizes with ITCH at plasma membrane and becomes ubiquitinated. In presence of antigen, localizes to immunological synapse at T-cell–APC contact area (peripheral and distal SMAC).
Background CXCR4 is a CXC chemokine receptor specific for stromal cell-derived factor-1. It has 7 transmembrane regions and is located on the cell surface. Acts with CD4 to support HIV entry into cells and is highly expressed in breast cancer cells. Mutations are associated with WHIM syndrome (warts, hypogammaglobulinemia, infections, myelokathexis). Alternate splice variants encoding different isoforms have been identified. [RefSeq, Jul 2008]
