Background
The protein encoded by this gene removes 5′ overhanging flaps in DNA repair and processes the 5′ ends of Okazaki fragments during lagging strand DNA synthesis. It interacts with AP endonuclease 1 during long-patch base excision repair, coordinating substrate transfer between enzymes. This protein belongs to the XPG/RAD2 endonuclease family and is essential for cell-free DNA replication. DNA secondary structures can inhibit flap processing at trinucleotide repeats, potentially leading to site-specific expansions.
